Site-specific Ultrasensitive Magnetic resonance of Mixtures for Isotopic Tracking

There is a high demand for the design of tools capable of tracking the origin of biological or synthetic molecules, in particular in the area of life, environmental, food and forensic sciences. Isotopic fingerprinting at natural abundance is the most promising way to address this challenge, thanks to Nuclear Magnetic Resonance (NMR) which is currently the only generic analytical technique that gives access to the site-specific isotope content. While the detection of very small relative differences between samples originating from different (bio)chemical pathways is possible through 13C isotopic NMR at natural abundance, this method can only be applied to simple and concentrated samples, owing to its low sensitivity. As a consequence, a large number of applications are currently out of reach of 13C isotopic NMR. To tackle this challenge, SUMMIT will develop a groundbreaking analytical workflow relying on two of the most powerful NMR methods: dissolution dynamic nuclear polarization (D-DNP) and ultrafast 2D NMR. This cutting-edge approach will allow the simultaneous measurement of isotopic fingerprints from multiple low-concentrated biomarkers molecules at natural abundance in complex mixtures, which is currently impossible with existing methods. The high potential of the resulting analytical strategy will then be demonstrated on a relevant biological study, ie. the investigation of the site-specific isotopic signature of breast cancer cells extracts, in order to identify (i) new biomarkers for the discrimination between cell lines expressing different hormonal receptors; (ii) novel potential therapeutic targets from the elucidation of metabolic pathways related to these biomarkers. As a result, SUMMIT will open new perspectives in the development of personalized medicine. Beyond this application, the project will have a high impact on a wide community of academic and industrial researchers, covering unmet needs from life sciences, food industry and forensic analysis.
