Résumé du projet

GENIUS aims to provide to French researchers and plant breeders state of the art genome editing tools in a variety of crop species.. 2012 Major event: Kick-off meeting 2013 Major events: Release of communication tools, first philosophical debate. Results: Design and production of site directed nucleases, choice of amiRNA backbones and production of respective constructs. First improvement of transformation rates of recalcitrant species (rose, apple). 2014 Major events: Meeting with scientific advisory board, project poster, signature of consortium agreement, second philosophical debate. Results: Production of stable transformants for nucleases, reporter constructs and landing pads. First evidence for nuclease-based deletion in tomato. Massive sequencing of land pad flanking sequences in rice and wheat. DNA cleavage (low efficiency) by a nuclease delivered as protein in moss. Improved transformation frequencies of certain elite lines (apple, rose). 2015 Major Events: Study day on ethics and food. Results: Demonstration of RAG1 activity in tomato. Functional NptII TALEN® in wheat open the way for gene targeting. Validation of miRNA backbones for gene extinction in rose and apple. Successful gene knock-out in rice (CRISPR/Cas9) and gene knock-in (gene editing) in tomato (TALEN®). Further optimization of rose regeneration, of rose and apple transformation and of protoplast regeneration in two elite potato varieties. 2016 Major Events: Departure of partner Cellectis, meeting with scientific advisory board, reorientation of work program, debate on urban food production, mapping of biotech projects. Results: Targeted DNA cleavage by RAG1 or I-SceI demonstrated in 5 crop species, landing pads established in 7 crop species. CRISPR-Cas9 mediated targeted mutations achieved in rice, maize and potato and CRISPR-Cas9 mediated gene editing in tomato. Delivery of CRISPR-Cas9 as ribonucleoprotein (RNP) complex in Physcomitrella. In planta transformation established for poplar.
